By demonstrating that the differentiation state and the fate of endothelial cells dictate the cellular response to PIK3CA mutations, we provide a novel paradigm for understanding the mechanisms dictating tissue-specific overgrowth in PIK3CA-related congenital disorders
Our findings expose the (i) generation of large clones in veins and capillaries causing vascular malformations, (ii) silent presence in arterial cells without pathological manifestations; and (iii) rewire of cell fate in arterial precursors
