Hi Oli,
FYI I advised a colleague to test your procedure on his protein and it deblocked the structure solving for him! It's a small protein also.
Great that you found this trick.

Just saw a talk about regulation of a Kir channel by a small RNA as well. But the mechanism of regulation is not clear.
Definitely not the two types of macromolecule we'd expect to interact...

All travelers from ESTA countries (yes the ones on visa waiver programs) will have to disclose 5 years of social media + huge amounts of personal data to enter the US now. All US academic associations should now meet outside the US if we want to meet our international colleagues.

Overall resolution is not the good metric to compare xray and cryo, but my guess is that local resolution cryo and overall xray are similar when you want to see holes in aromatics.
At these resolutions, diffraction is mostly isotropic and treated as overall for the whole dataset, but not cryo.

AQuaRef: machine learning accelerated quantum refinement of protein structures - Nature Communications AQuaRef employs machine learning to refine protein structures from cryo-EM and X-ray data in Phenix. Finally out in print! Long time project with @olexandr.bsky.social and the PHENIX group. Refinement of X-ray and CryoEM structures, using Machine Learned Potentials as the back end to help fitting. This fulfills the old dream of doing QM refinement, but very very cheap www.nature.com/articles/s41...

Looking very promising, it's a great idea. Can't wait to test it.

Thanks for sharing Oli.
FYI I tried it on a larger membrane protein for which the 2D classes were not high res (not enough particles) but I hoped for a miracle. The miracle didn't come, I ended with a high res bad model, a negative control I guess.
I'll nevertheless keep the preprint in my folder

Soon... a new building for our laboratory, with 1400m² extra and renovated space to host new equipements such as cryoEM, CryoFIB-SEM, optical microscopies in biosafe confined environment, and a synthetic biology labs.

This one is a bit of a departure from the usual and definitely a work in progress!

We found that by using ab initio reconstruction at very high res, in very small steps, we could crack some small structures that had eluded us - e.g. 39kDa iPKAc (EMPIAR-10252), below.

AlphaFold developer Google DeepMind to fund CASP as NIH funding falls short Protein structure prediction contest CASP gets temporary funding from Google DeepMind as NIH grant runs out.

NEW from me at STAT:

We've learned that DeepMind is going to fund CASP, the protein structure competition that brought Google DeepMind's #AlphaFold to prominence (and a Nobel Prize), as its NIH funding runs out.

Tucked away in the 'One Big Beautiful Bill' is the requirement that all visitors to the US must pay $250 just to enter the country starting 1 October! For a family of 5 that's $1250! I doubt this will help America's crashing tourism business which is down 10% already since January

We are happy to release LocScale2.0: a tool for context-aware, confidence-weighted cryoEM map optimisation.

Taking two half maps as input, LcoScale-2.0 produces feature-enhanced maps along with a robust confidence score that guides objective map interpretation.
