Why it matters:
This pDC → cDC plasticity could let us tune immunity — boosting antiviral defenses or damping harmful inflammation in autoimmunity.

This isn’t just in a petri dish:
We saw it in human skin inflammation, wounds, and even healthy aging.
pDC plasticity happens in vivo — actively shaping immune responses in health & disease.

Using single-cell omics + functional assays, we saw activated pDCs lose IFN-I machinery & gain cDC type 2 (cDC2) traits— a full fate switch.

Two key regulators:

TNF → is the switch push
IFN-I → is the restrain

pDCs are famous for cranking out type I interferon to fight viruses.
Conventional DCs (cDCs) present antigens to activate T cells.
But here’s the puzzle: why are pDCs part of the dendritic cell family at all? 🤔
Turns out, they can change teams.
