Our findings highlight region-specific differences in longitudinal rsFC strength between β€œnormal” aging and early AD pathology and shed light on early changes in rsFC strength with AD pathology, which might already be disadvantageous for episodic memory performance in APOE4 carriers.

Longitudinal increase in rsFC strength between the anterior hippocampus and the superior precuneus is associated with higher early AD pathology. Higher rsFC strength was related to cognitive decline only in APOE4 carriers.

Longitudinal decrease in rsFC strength within the posteromedial cortex and between the posterior hippocampus and the inferomedial precuneus was associated with β€œnormal” aging. Pathology-independent lower rsFC strength was related to cognitive decline.

In essence, hypoconnectivity was related to aging, hyperconnectivity was related to early Alzheimer’s pathology.
But which specific regions were involved?!

We investigated resting-state functional connectivity (rsFC) in episodic network regions over two years in cognitively normal amyloid- and tau-negative older adults (β€œnormal” aging sample) and in older adults with CSF measurements (AD-biomarker sample).

Differential effects of aging, Alzheimer's pathology, and APOE4 on longitudinal functional connectivity and episodic memory in older adults Both aging and Alzheimer's disease (AD) affect episodic memory networks. How this relates to region-specific early differences in functional connectivity (FC), however, remains unclear. We assessed re...
