Neurodegenerative diseases need more mechanism-informed trials - Nature Medicine Recent failures in high-profile clinical trials of neurodegenerative diseases highlight the need to rebalance clinical ambition and biological understanding.

"Mechanistic sufficiency needs to be considered a priority before a therapy enters clinical trials. (...)" - Will a detailed dissection of pathways and mechanisms of neurodegeneration regain broader appreciation?

Ultrastructural membrane dynamics of mouse and human cortical synapses Eddings et al. illustrate that synaptic membrane trafficking can be studied with millisecond resolution in resected human brain tissues using the technique “zap-and-freeze” time-resolved electron microscopy. Using this method, they find that synaptic vesicles are recycled via ultrafast endocytosis in human neocortical slices.

Evidence that mitochondria in macrophages are destroyed by microautophagy - Nature Communications Mitophagy, the selective degradation of mitochondria, has been thought to occur via macroautophagy. Here, the authors present data indicating that microautophagy also mediates mitophagy in macrophages, and that it is essential for immune activation during M1 polarization.

And this on top! www.nature.com/articles/s41...

The emerging mechanisms and functions of microautophagy - Nature Reviews Molecular Cell Biology Microautophagy involves direct engulfment of cytoplasmic components, including proteins and organelles, by lysosomes and late endosomes for degradation. Although it is one of three main types of autophagy — along with macroautophagy and chaperone-mediated autophagy — its mechanisms and physiological roles have only recently begun to emerge.

www.nature.com/articles/s41...
Excellent review! There is mounting evidence that results based solely on rapamycin and LC3 are insufficient to conclude that the observed changes are related to macroautophagy.

How do neuronal lysosomes maintain functional synapses? - BIOspektrum The complex architecture of neurons poses unique challenges in maintaining proteome balance. Lysosomes, cellular metabolic hubs and main degradative organelles, play a central role in this process, thus supporting synaptic activity. Recent findings suggest that a specialized subpopulation of lysosomes in dendrites fuse with plasma membrane and release aggregation-prone proteins to the extracellular space. At the same time, they lead to structural changes in synapses, an intriguing link between the control of protein concentration and structural synaptic plasticity.

doi.org/10.1007/s122...
Lysosomes are much more than 'garbage dump', they are metabolic hub with really cool functions! If you are interested in this topic but would not like to go to deep in the molecular pathways (which are cool as well!), I hope you may find this contribution useful!

Other thing is that I have reviewed recently a paper with RNA seq data and the file with the seq list was missing. While some journals do request it in the publication phase, I believe that the editors should not send such a manuscript to review. It is incomplete.

Oh, I am sorry to hear that, this is really wrong. I do not have such an experience but actually I have never tried (I so far requested data from collaborators only). Would be cool to be able to initiate official data request via a link on the journal website.
