we observed unexpected responses in melanoma patients with driver mutations in HRAS, NF1, and GNAQ, indicating that exceptional responses to combined T-VEC and binimetinib may extend beyond NRAS-mutated melanoma.

#Intratumoral #CD4Tcells following combination therapy were enriched for a CXCR6+ Th1 effector subset distinct from the follicular helper (Tfh) phenotype associated with tumor-reactive CD4+ T cells

6 of 7 patients (85.7%) who received #TVEC and binimetinib therapy achieved a complete response compared to 2 of 7 (28.6%) patients with NRAS-mutated melanoma treated with T-VEC alone or combined with #checkpoint blockade agents

we find that #TVEC combined with #MEK #inhibitor #binimetinib induced exceptional clinical responses by amplification of pre-existing #Tcell responses and induction of de novo tumor-reactive immunity.
