Fast and Luminous: Clip-tag2. As fast as Halo and SNAP-tag2 with some benefits of redesigned substrate, PF instead of BC is from now on CLIP team. We look forward seeing cool applications in multiplexing, CID, sensors, and maybe some new surprising ones!

A Cinderella story: raising CLIP-tag from the ashes to shine in fluorescence labeling.

Great work, and congratulations Stephan and the whole team. I can not skip asking about submission date in 2021. What happened in the meantime that they kept it on and off so long?

SNAP-tag2 for faster and brighter protein labeling - Nature Chemical Biology SNAP-tag is a widespread tool for labeling protein for bioimaging. Now, Kühn et al. report SNAP-tag2 with increased labeling kinetics and brightness, which translates into a better performance in live...

📣Paper alert! Interested in faster and brighter protein labeling than before? 👀 @steffi-k.bsky.social & other colleagues have developed an improved tool for labeling proteins with synthetic fluorophores for bioimaging. Also works in STED microscopy 🤩🔬 Congratulations!🥳

SiR-XActin, SPY650-Fastact and SiR-actin, along with whole jasplakinolide-like actin ligand based tools are cell permeable small molecules which do not require genetic manipulation. Dissolve in DMSO, add some to the medium and image with your favourite microscope :)

Can you spot the difference? 🔬
SiR-actin labeling can be cytotoxic and affects actin dynamics - while the new XActin probes do not! Developed by @veselin-nasufovic.bsky.social and perfect for time-resolved and STED imaging without the need for genetic manipulation.

New actin probe from @veselin-nasufovic.bsky.social et al! SiR-XActin and its variants are more photostable and less perturbative than SiR-actin or FastAct. And it's already available from @spirochrome.com! With some beautiful data from @chillinwithpfn1.bsky.social:

We hope that #SiR-XActin will continue the legacy of older relative #SiR-actin and become an indispensable tool in live-cell no-wash imaging, cell painting assays and cell biology in general.

4/5 🔬 We have demonstrated that #𝗫𝗔𝗰𝘁𝗶𝗻 spectral properties can be tuned with other #SiR and #MAP fluorophores covering the important part of the imaging window.

3/5 💡 The key to our success was a careful #cryo-EM structure-driven SAR study of the #XActin ligand, in which we synthesized and tested more than 100 cycic peptides. We minimized cytotoxicity and preserved its binding to F-actin.

2/5 🌟 We've developed a new fluorescent probe for live-cell no-wash imaging with enhanced photostability and no observable cytotoxicity. 𝗦𝗶𝗥-𝗫𝗔𝗰𝘁𝗶𝗻 enables imaging of actin dynamics without induction of non-physiological phenotypes.
