Finally, we map initial DNA unwinding genome-wide at near base-pair resolution. We found that strand separation initiates at the interface between opposing MCM2-7 hexamers, capturing this transient step of helicase activation in vivo for the first time.

By systematically probing all MCM2-7 interfaces in vivo, we identify the Mcm2/5 interface as a unique ssDNA exit gate during helicase activation. This gate extrudes ssDNA as the inactive double hexamer converts into two active CMG helicases while not interfering with ongoing DNA synthesis.

Using a rapamycin-inducible linkage system in living yeast, we show that blocking rotation and separation of the two MCM2-7 hexamers specifically delays release of the Sld3 loading factor, revealing a direct link between helicase mechanics and activation-complex disassembly.
