TurboCas: A method for locus-specific labeling of genomic regions and isolating their associated protein interactome Cenik et al. describe TurboCas, a single-guide-RNA-directed dCas9-miniTurbo construct that can identify chromatin-binding proteins at single loci in the mammalian genome. Labeling of the FOS promoter identifies heat shock regulators, including FUBP3. MYC promoter labeling demonstrates the super elongation complex and BRD4 as MYC regulators.
