Another thing that may work for you is to keep the tmp directory as C:\tmp and manually deleting the _gbsbuild, _gbscache and _gbstools folders inside that, or clicking "Empty Build Cache" in the UI which does the same. I saw a similar issue to you on Windows just now, might need a bug fix release.
It's been a while but I seem to remember Windows didn't like having long paths for temporary directory and I had to just revert to using C:\tmp in cases where you provide too long a file path. Could you maybe try something shorter that you have permission to write to e.g. something like C:\gbstmp
It looks like your user doesn't have permission to write to the temporary folder GB Studio uses anymore.
A quick fix would be to go into application settings and set a different temporary directory to somewhere you have permissions on your computer (screenshot is Mac but it's the same on Windows)
It's been over a year since the last major update but I hope it's been worth the wait!

LGBTQ+ focused game narratives that use the artstyle and mechanics of a retro Game Boy game. All of these can be found on @itch.io and most are made with @gbstudio.dev which means you can even play them on a physical Game Boy.
Working on some tools to help diagnose slowdown in @gbstudio.dev games. Can scrub through recordings and see what was running each frame. Already found a few easy wins for optimisation coming to the next release! Code up at github.com/chrismaltby/... if anyone might find it useful (plugin devs?)
Finished adding splash effect for when the hero falls into the water. I think it came out well.
