An amazing preprint by @gerlichlab.bsky.social on the fate of DNA during apoptosis. Definitely to add on your reading list!

Our SPY650-DNA probe was heavily used to visualize DNA throughout the manuscript.

Congratulations to the team!

Hello epithelial mechanics fans!! I’m Juanma @juanmagararc.bsky.social πŸ‘‹ I work on cell mechanics (see celldynamicslab.com) and use Flipper-TR FLIM to probe membrane biophysics in cells.

Join me on this tour about Flipper: what it measures, strengths, advice, cool case studies, and cute drawings!

You are correct! mtDNA staining is typically performed with an intercalating DNA dye with nanomolar affinity for dsDNA. SPY-DNA probes, primarily designed for live cell imaging, are DNA groove binders and have a lower affinity to avoid toxicity. Probably not strong enough for robust mtDNA staining.

The images shows an array of 6 Expansion microscopy images of cell nuclei (three columns and 2 rows). each image was taken with a different SPY-DNA probe to stain the nuclear DNA and is pseudocolored to the fluorescence wavelength of the SPY-DNA probe. Trom top left to bottom right: SPY505-DNA (green); SPY555-DNA (magenta); SPY595-DNA (yellow); SPY620-DNA (orange); SPY650-DNA (red); SPY700-DNA (darker red). a white scale bar of 50 micrometers is given for each image to get an impression of the scale of each nuclei.

Are you staining DNA in your ExM samples?

We have awesome news for you! All our 6 SPY-DNA probes work beautifully well with the U-ExM protocol.

You can now tune the color of the DNA channel to your needs, according to the fluorophores on your preferred antibodies.

Peroxi_SPY probes were designed for peroxisome live cell imaging. Turns out they work with fixed cells as well!
Peroxi_SPY can thus be combined with immunofluorescence labeling.

A new article provides a detailed protocol: app.jove.com/t/69405/pero...

It turns out that, in addition to its very low phototoxicity, PKmito Deep Red (PKMDR) directly reports on mitochondrial membrane potential (MMP) in live cells through its lifetime!
